چکیده

Introduction Oxidative stress is an important part of a wide range of pathologies and therefore an interesting parameter to determine. However, the detection of reactive oxygen species (ROS), is not straightforward. Sample heterogeneity, delayed analysis and sample preparation, reduces specificity and sensitivity due to probe activation by light, air, probe leakage from cells or cellular activation by sample manipulation. Hence, samples should be processed minimally and analysed immediately if possible. Since clinical research can be subject to uncontrollable timetables and only few departments have access to a dedicated laboratory, the quantification of ROS is challenging.

Role of Caspases and Reactive Oxygen Species in Rose Bengal-Induced Toxicity in Melanoma Cells

Objective We have previously shown that Rose Bengal (RB) alone, not as a photosensitiser, could induce apoptotic- and non-apoptotic cell death in different melanoma cell lines. To clarify RB-induced toxicity mechanisms, role of caspases and reactive oxygen specious (ROS) were studied in melanoma cells. Material and Methods Human melanoma cell lines, Me 4405 and Sk-Mel-28 were cultured in DM...

Detection of defective granulocyte function with flow cytometry in newborn infants.

Reactive oxygen species (ROS) production by phagocytic leukocytes is a critical factor in immunity against microorganisms. Human neonates are susceptible to overwhelming infections, for which abnormal granulocyte function may play an important role. In this study, we aimed to identify a convenient and quantitative method to measure ROS production by human granulocytes after cellular activation....

Effects of lidocaine hydrochloride on canine granulocytes, granulocyte CD11b expression and reactive oxygen species production.

Lidocaine hydrochloride (Lido) is widely used for analgesia in veterinary medicine; however, in humans, it has been suggested that Lido attenuates granulocyte functions, such as adhesion and reactive oxygen species (ROS) production. Thus, Lido may affect canine granulocyte function; however, there have been no reports on the effects of Lido on canine granulocyte function. Thus, we studied the e...

Protective effect of Viola tricolor and Viola odorata extracts on serum/glucose deprivation-induced neurotoxicity: role of reactive oxygen species

Objective: Oxidative stress plays a key role in the pathophysiology of brain ischemia and neurodegenerative disorders.Previous studies indicated that Viola tricolor and Viola odorataare rich sources of antioxidants. This study aimed to determine whether these plants protect neurons against serum/glucose deprivation (SGD)-induced cell death in an in vitro model of ischemia and neurodegeneration....

Use of chemical probes to detect mitochondrial ROS by flow cytometry and spectrofluorometry.

Aerobic respiration is a major source of energy in eukaryotic cells. In this setting, ATP production by the mitochondrial respiratory chain relies on the availability of NADH and FADH2 to donate protons and electrons. The flux of electrons down the electron transport chain, based on a series of oxidation-reduction reactions, releases energy that allow for the transport of H(+) ions across the i...
